Abstract: Chromatography is an analytical technique based on the separation of molecules due to differences in their structure and/or composition. In this article, the author discusses practical aspects of high-performance liquid chromatography theory (chromatographic principles such as retention, resolution and sensitivity), chromatographic mechanisms, instrumentation, mobile phase and reservoir, pumps, injectors, detectors, data-acquisition/display systems and columns, separation techniques (isocratic vs. gradient elution and derivatization), quantitative analysis (external standard and internal standard) and validation. Figures show a chromatogram of a reference standard and a sample. High-performance liquid chromatography is currently the most widely used method of quantitative analysis in the pharmaceutical industry and in pharmaceutical analytical laboratories.
